Dear Roivant family,
Today Axovant announced topline results from its phase 3 MINDSET trial. Unfortunately, intepirdine failed to achieve a statistically significant result on the primary endpoints of the study. I have attached Roivant’s and Axovant’s public statements on the trial outcome. Rather than repeating our public remarks, I would like to share some candid reflections with you about what today’s announcement means for all of us. I do not intend to write a letter like this every time we complete a major clinical study, but I wanted to share my thoughts today because MINDSET was the first major phase 3 study completed by a company in the Roivant family. I also look forward to speaking with you during the Vant townhall at 9am EST.
First, we are grateful for the contributions of our staff, Axovant’s staff, our investors, our partners, our scientific investigators and advisors, and most importantly the trial participants and their loved ones. We are committed to making sure everyone’s contributions to MINDSET were not in vain, even though the trial itself did not succeed.
There is no doubt that the MINDSET failure is a major setback for Axovant, but it is not the end of the road. First, the most important ingredient for the success of any company is the quality of its people, starting with its leadership team. I am confident that there is no one on the planet better suited to lead Axovant right now than its CEO David Hung. Over the decade that I have known David, I have admired not just his ability to build Medivation into one of the most successful biotech companies in history, but the fact that he did so after the failure of his lead drug in Alzheimer’s disease. He overcame that setback and harnessed his team to rebuild Medivation into an even greater company than anyone imagined at the time. I am fully confident in David’s ability to do the same, if not more, at Axovant.
Axovant’s commitment to the field of dementia, and neurodegeneration more broadly, remains as strong as ever. Axovant is continuing to explore future opportunities for intepirdine in dementia with Lewy bodies, as well as the rest of its pipeline. And Axovant will continue to build its pipeline through strategic in-licensing and acquisition of new promising drugs in the neurosciences.
Roivant does not view – and has never viewed – Axovant as simply a ‘vehicle’ for developing intepirdine, but instead as a platform for the development of high-impact drugs in dementia and the neuroscience field more generally. Intepirdine itself may have a future at Axovant, but Axovant’s future as a company will rest on more than just intepirdine.
A reasonable question on your mind may be how the MINDSET failure affects the overall Roivant business model. Let me reiterate what I have shared with all of you in the past: the impact of today’s announcement will be slight at most. Roivant’s future has always hinged on much more than the success or failure of any one of our clinical programs. I can confirm that this remains the case today: we will continue to aggressively pursue our long-range mission of reshaping the drug development process for the better. We have 15 other drugs in the pipeline and a team that I believe is second to none in its ability to identify promising drugs in new therapeutic areas, and to launch more Vants around those drugs. And crucially, we have the resources that we need to build upon that foundation.
On the back of today’s announcement, I fully expect many outside observers will question our strategy of pursuing the development of shelved or discontinued pipeline drugs. I don’t fault those who would ask the question. After all, we truly believed in the development of intepirdine in the MINDSET trial, and it clearly did not succeed. I have also long held that Roivant’s metric of success over the long run will be how successful we are in getting new medicines to patients, and we are still yet to accomplish that long-range goal. But I am confident that over time we most certainly will.
Even following the results of MINDSET, I still believe it was unambiguously the right decision for us to take the bet on intepirdine. The scientific rationale was sound, and the potential benefit to society was large. Those are exactly the kinds of bets we want to make. We have other promising drugs in development at Roivant, and we will not shy away from taking bold bets in the future as well.
In my opinion, in addition to the success we have achieved in launching new Vants and in pursuing development of a variety of drugs, one of our most important achievements has been the growth of Roivant itself, as a platform to support the efficient identification and advancement of medicines across all of the companies in the Roivant family. I could not be more proud of every one of you who has helped us get here. The caliber of talent at Roivant and across the Vants today is as high as anywhere I have ever seen: you are some of the most brilliant, passionate, and driven people I have ever met in my life.
Today’s failure is challenging and humbling for me on a deeply personal level – not because it dampens my confidence in Roivant’s business model, and not because I have any concerns about Roivant’s long-range prospects for success. Indeed I have no doubts about Roivant’s bright future. Rather, it is painful and disappointing because the success of MINDSET would have been so impactful to so many. It is simply impossible for me to be dispassionate about it. While this is personally difficult for me, that may not be a bad thing for our business: I will be sure to harness the ‘sting’ I feel now to double down on my efforts to ensure that we succeed as a company, and that Roivant will be even stronger for having dealt with the experience of failure. I am certain that we will achieve our goal of improving the process of getting new medicines to patients. It is only a question of ‘when,’ not ‘if.’
I am deeply grateful for your hard work, your confidence, and your commitment. Stay focused. Stay scrappy. We have much to accomplish.
